Title:
Development of Ultrasound Theranostics System

Abstract:
“Theranostics” is a treatment strategy that combines therapeutics with diagnostics. Ultrasound is a good tool for “theranostics” due to have multi-potency both of diagnostics with sonography and therapeutics with high intensity focused ultrasound (HIFU). Microbubbles and nanobubbles are utilized as not only contrast imaging agent but also enhancer of drug and gene delivery. However, it has not developed active targeting bubbles and effective bubbles for delivery.
Liposome can be modified with active targeting moiety and encapsulated various molecules. Therefore, liposome technology is very important to develop an ideal drug and gene delivery system. Thus, we attempted to make an intelligent nanobubble with liposome technology, and developed novel liposomal nanobubble (Bubble liposome (BL)) containing ultrasound imaging gas, perfluoropropane. In addition, BL modified with RGD peptide which binds to a v b 3 integrin had potency as imaging agent for blood clot. When the accumulated BL into the clot was exposed to therapeutic ultrasound, cavitation was induced and resulted in disrupting the clot. It is suggested that this would be unique clot imaging and therapeutic system. Moreover, we have reported that the combination of BL and ultrasound is an effective drug and gene delivery system. Therefore, it is expected that our system would be an important technology to establish an ideal “Theranostics”.
